Abstract [eng] The impact of chemical and physical factors on the germination of Geobacillus stearothermophilus spores in the agar media was studied. For this purpose chemical agents such as carbohydrates, amino acids, inorganic salts, adenosine and lysozyme were used. Spore germination differed at various concentrations of the same carbohydrate. G. stearothermophilus spore germination was activated by a 1 % lactose, 1 % soluble starch and 0.5–1 % sacharose. Various amino acids differently affected G. stearothermophilus spore germination. Cysteine concentrations (0.01–0.2 %) did not affect spore germination, but the spore count at DL-arginine concentrations varying from 0.01 % to 0.2 % was significantly higher. L-alanine (0.03 %) activated germination of G. stearothermophilus spores, while DL-alanine (0.03 %) did not have any impact on the spore growth. The effects of inorganic salts showed that spore germination was stimulated more intensively (by 50 %) compared to control medium by adding 0.4 % KH2PO4, 0.2 % K2HPO4, 0.03–0.05 % NaHCO3 and 0.03 % NaNO2. It was found that concentrations of ZnSO4 and CoSO4 (0.005–0.04 %, resp.) inhibited metabolic reactions of G. stearothermophilus spores and prevented from germination. FeSO4, K2SO4 and NaCl did not have any impact on spore germination because significantly lower spore counts were obtained on the agar media with these agents than in control medium. Spore activation was influenced by thermal treatment as a physical factor. It was found that spore germination was optimal when the spore suspension was treated at 70 oC for 5 min and was immediately plunged into an ice bath.
