Abstract [eng] Aim of work: evaluate proton pump inhibitor lansoprazole effect on doxorubicin and pegylated liposomal doxorubicin (PLD) transport into breast cancer cell cultures. Objectives: 1. Investigate doxorubicin and PLD penetration into MCF-7 breast cancer cells. 2. Evaluate proton pump inhibitor lansoprasole effect on doxorubicin and PLD transport into MCF-7 breast cancer cells. 3. Examine doxorubicin and PLD transport into breast cancer MCF-7 cell spheroids. 4. Evaluate doxorubicin and PLD transport into breast cancer MCF-7 cell spheroids after incubation with lansoprazole at different pH values. Experimental methods: In this work we have evaluated lansoprazole effect on doxorubicin and PLD penetration into breast cancer MCF-7 cell cultures. Toxicity of the compounds was evaluated by MTT assay. The penetration of doxorubicin and PLD into cells and spheroids in a slightly basic (pH 7.2) and acidic (pH 6.0) medium was evaluated by fluorescence microscopy. The penetration of doxorubicin or PLD transport into cells and spheroids was evaluated after lansoprazole prieincubation for 2 hours before adding doxorubicin and PLD. Photos were analysed using ImageJ software. Results: In a slightly basic environment (pH 7.2) doxorubicin penetration into the cell nuclei was better after 2 h of incubation. After longer periods of incubations, it‘s penetration remained the same. In slightly acidic environment (pH 6.0) doxorubicin penetration into the cell nuclei and the whole cell increased constantly during the whole 4 h incubation. Before the incubation with doxorubicin, cells were prie-incubated with lansoprazole at both slightly basic and acidic media. Cellular uptake of doxorubicin after 4 h of incubation were 4 times greater than in the control group. PLD penetrated better into the cells at slightly basic environment, although lansoprazole has no influence on penetration of PLD at either slightly basic or acidic pH compared to the control group. The penetration of doxorubicin was dependant on the time of incubation. Lansoprazole enhances doxorubicin penetration at slightly basic and slightly acidic environment. Lansoprazole doesn‘t increase PLD cellur uptake into the tumor spheroids.
