Abstract [eng] In biotechnological processes, the aim is to maximize product yield. Direct measurement of product concentration in bioreactors is costly. To address this issue, biotechnological processes utilize sensorbased systems supported by mathematical, statistical, or hybrid models. These systems continuously monitor process parameters and utilize the data to estimate concentrations of product and other process components in real-time. This paper describes a mathematical model developed to estimate real-time concentrations of biomass, lactose, and recombinant protein in a fed-batch E. coli process bioreactor. The mathematical model applies equations for oxygen consumption rate in the bioreactor to estimate concentrations of biomass and recombinant protein. The paper also discusses parameter identification functions, which train the mathematical model based on experimental data and enable more accurate estimation of component concentrations in the bioreactor by the sensor-based system. The mathematical model was evaluated using data from four experiments, identifying parameters for different experiment sets and training the model to estimate concentrations based on more than one experiment.
